FACSAria II

Flow cytometry sorter with FACSDiva software

The Aria II (Upgraded in Fall of 2008) is high speed cell sorter that can sort up to 70,000 events per second and can analyze up to 15 parameters.  The IU Bloomington Aria II is a custom build unit that includes:

  • 4 lasers: Blue – 488nm, Red – 633nm, Violet – 407nm, and UV – 355nm. 
  • 2 nozzles sizes: 70um, 85um, 100um, and 130um
  • Aerosol management system
  • Temperature control system
  • 2 and 4 way sorting
    • 2 way sorting into 5mL or 15mL tubes
    • 4 way sorting into 5mL or 1mL tubes

It has an Automated Cell Deposition Unit (ACDU) designed for sorting into multiwell plates or onto microscope slides.

LSRII

Flow cytometry analyzer with FACSDiva software

The LSRII is a 3 laser analyzer, capable of multicolor analysis of up to 8 colors.  The LSRII includes the following:

  • 3 lasers: Violet - 405nm, Blue – 488nm and Red – 633nm
  • Sample injection port (SIP) diameter: 150um

FACSCalibur

Flow cytometry analyzer with CellQuest Pro software

The FACSCalibur is a 2 laser analyzer, capable of multicolor analysis of up to 4 colors.  The FACSCalibur includes the following:

  • 2 lasers: Blue – 488nm and Red – 633nm
  • Sample injection port (SIP) diameter: 150um
  • Sort option (low speed)

COPAS Select

Flow cytometry large particle sorter

The COPAS Select is a large particle sorter, which in highest sort mode, can sort from 10-15 events per second.  It can sort C. elegans, Drosophila, plant seeds and cells, and beads, and can perform multicolor analysis of up to 3 colors.

  • 1 laser: Blue – tunable to 488 or 514nm excitation, detects Green, Yellow, and Red emission wavelengths (510, 545, and 580nm)
  • Flow cell size:  500µm – can collect events from 20-350µm in diameter
  • 1 way sorting: into collection device
  • X-Y stage: for sorting into 24 or 96-well plates

AmpliGrid

Single-cell PCR system

The AmpliGrid system (added to the flow facility as of February 2009) is a single-cell PCR system that can be used in conjunction with the FACSAria II cell sorter.  The AmpliGrid slide contains 48 reaction spots where single cells may be deposited.  The slide is then placed on the AmpliSpeed thermal cycler, 1μl of mastermix are added and each spot covered with 5μl of immersion oil.  

Applications include:

  • PCR
  • RT-PCR
  • qPCR*
  • Sequencing

Cost for usage:  There is no cost for usage of the AmpliSpeed cycler.

AmpliSpeed cycler training is available and users may self-run this instrument after a short training session.  Alternatively, users may have our dedicated technician run samples for them.

*Please note, when performing qPCR, the AmpliGrid and AmpliSpeed are designed to complete only the reverse transcription step of the qPCR protocol.  Final amplification and analysis can then be performed on another system such as the Stratagene Mx3000P Quantitative PCR Thermal Cycle (available at the Center for Genomics and Bioinformatics - MY162). 

 

Z2 Coulter Counter

The Z2 instrument is a cell and particle counter that reports both cell counts and concentration of a cell population, as well as size distribution of that cell population.
